    描述:
    重组人碱性成纤维细胞生长因子(bFGF)由146个氨基酸组成的相对分子量为16kD的多肽。采用基因工程技术和独特的色谱技术分离纯化、冻干制备。bFGF具有强有力的促进细胞分裂增殖的生物活性。

    用途:
    bFGF通过与细胞膜上的受体,广泛作用于外胚层、内胚层和中胚层多种来源的细胞,包括上皮细胞、粘膜细胞、内皮细胞、平滑肌细胞、骨骼肌细胞、卵巢颗粒细胞、成纤维细胞和神经细胞等,都具有促丝裂原作用和趋化作用,是一种与组织创伤的愈合过程密切相关的内源性活性因子。主要作用机制为:
    l)趋化作用:rh-bFGF作为化学趋化剂趋化多种炎性细胞、组织细胞与修复因子,向创面或伤口迁移,为创面杀菌以及后期修复创造重要和必需的条件。
    2)增殖作用:①rh-bFGF直接作用于组织修复细胞上的生长因子受体,通过其促分裂效应加速细胞周期转变来加速创面修复;②rh-bFGF通过竞争性作用上调组织修复细胞上生长因子受体的活性,从而加快信号的传递。
    3)重建作用:bFGF通过增加细胞内胶原蛋白、连接蛋白、透明质酸、羟脯氨酸、糖蛋白的合成,提高各种再生细胞间的连接强度和修复质量,减少和预防疤痕的形成。
    应用领域:
    作为生物功效性原料添加以下产品中使用。
    ① 抗皱产品:激活细胞代谢,促胶原纤维合成,达到抗皱效果;
    ② 修复产品:促进皮下神经和血循环组织的再建,增强修复效果。

    Description:
    Basic fibroblast growth factor possess broad mitogenic and cell survival activities,and are involved in a variety of biological processes, including embryonic development、cell growth、morphogenesis、tissue repair、tumor growth and invasion. Recombinant Human Basic Fibrobast Growth Factor produced in P.pastoris, polypeptide chain containing 155 amino acids and having a molecular mass of 17353 Dalton.The Bfgf is purified by gene techniques and proprietary chromatographic techniques.

    (a)        Analysis by RP-HPLC.

    Amino acid sequence:
    The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Glu-Gly-Glu-Ile, which agrees with the sequence of native human bFGF.
    Biological Activity:
    The ED50 calculated by the dose-dependant proliferation of BAF3 cells expressing FGF receptors is less then 0.5ng/ml corresponding to a specific activity of 2×105 Units/mg.

    Storage:
    Lyophilized Epidermal Growth Factor although stable at room temperature for 18 days; and stored at 2-8°C for 3 years.
    Usage:
    Through the cell membrane receptors , bFGF extensively acts on ectoderm, endoderm and mesoderm cells of a variety of sources, including epithelial cells, mucosal cells, endothelial cells, smooth muscle cells, skeletal muscle cells, ovarian granulosa cells, fibroblasts and nerve cells and so on, have a role in promoting mitogen and chemoattractant, is a endogenous factors which is closely related to the tissue healing process.The main mechanism is as following:
    l) Chemotaxis:As a chemoattractant, rh-bFGF drives multiple inflammatory cells, tissue cells and repair factor towards  the wounds or the injury ,creating important and necessary conditions for wound sterilization and late repair.
    2) proliferation:①rh-bFGF directly acts on the growth factor receptor located on tissue repair cells, through its mitogenic effect to accelerate cell cycle changes to accelerate wound healing; ②rh-bFGF through competitive effect increase the activity of growth factor receptor located on tissue repair cells, sequentially accelerating the signal transduction.
    3) reconstruction: bFGF by increasing the synthesis of intracellular collagen,fibronectin,hyaluronic acid,hydroxyproline and glycoprotein enhances the connection intensity and repair quality among various renewable cells result in reducing and preventing scar formation.
